3DIANA

3DIANA analyzes protein domain–domain interactions and supports quaternary structure modeling by integrating electron microscopy (EM) maps with structural data from X-ray crystallography and NMR.

Key Features:

  • Integration of Structural Data: Combines medium- or low-resolution electron microscopy maps with structural information from X-ray crystallography and NMR to support hybrid structural modeling.
  • DIMERO Interaction Scoring: Applies the DIMERO method to statistically evaluate potential domain–domain contacts by scoring physical binding sites based on protein interaction network topology.
  • Domain Interaction Catalog: Provides a catalog of structurally resolved and predicted interacting regions involving protein domains.
  • Structural Template Identification: Suggests structural templates for modeling interactions between protein domains when direct experimental structural data are limited.

Scientific Applications:

  • Protein Interaction Network Analysis: Identifies and evaluates potential domain–domain contacts to study molecular interaction networks.
  • Quaternary Structure Modeling: Supports modeling of protein complex architectures using predicted and experimentally derived interaction information.
  • Hybrid Structural Modeling: Integrates EM maps with X-ray crystallography and NMR data to construct hybrid models of protein assemblies.

Methodology:

3DIANA integrates electron microscopy maps with structural data from X-ray crystallography and NMR and applies the DIMERO method to score potential domain–domain interactions based on protein interaction network topology.
