CRISPRdirect

CRISPRdirect identifies optimal CRISPR/Cas9 guide RNA (gRNA) target sites to enable specific genome editing by minimizing off-target effects.

Key Features:

  • gRNA selection: Identifies optimal guide RNA (gRNA) target sequences that meet stringent criteria for effective and precise CRISPR/Cas9 editing.
  • PAM filter (5'-NGG): Requires a 5'-NGG protospacer adjacent motif immediately following the target site.
  • Uniqueness/off-target check: Assesses uniqueness of target sequences within the entire genome to reduce potential off-target interactions.
  • Exhaustive genomic searches: Performs exhaustive searches against an extensive genomic sequence database covering human, mouse, rat, marmoset, pig, chicken, frog, zebrafish, Ciona, fruit fly, silkworm, Caenorhabditis elegans, Arabidopsis, rice, Sorghum, and budding yeast.

Scientific Applications:

  • Genome engineering: Designing specific CRISPR/Cas9 target sites for genome editing projects.
  • Basic biological research: Enabling targeted perturbations for functional studies in diverse model organisms.
  • Applied biotechnology: Supporting development of biotechnological innovations that require precise genetic modifications.
  • Cross-species gRNA design: Tailoring gRNAs for multiple species by leveraging comprehensive genomic coverage.

Methodology:

Performs exhaustive searches against genomic sequence databases and applies filters requiring a 5'-NGG PAM immediately following the protospacer and sequence uniqueness across the entire genome to minimize off-target effects.
