EvoPPI
EvoPPI integrates and compares protein–protein interaction (PPI) datasets from BioGRID, CCSB, DroID, FlyBase, HIPPIE, HitPredict, HomoMINT, INstruct, Interactome3D, mentha, MINT, and PINA2 to identify orthologous proteins and conserved interactions across species using a BLAST-based approach.
Key Features:
- Cross-Species Comparison: Uses a BLAST-based approach to identify orthologous proteins and enable interspecies PPI comparisons despite differences in protein nomenclature.
- Interactome Analysis: Performs comprehensive analysis of interactomes for individual proteins or protein groups to characterize their functional networks.
- Database Integration: Consolidates PPI data from BioGRID, CCSB, DroID, FlyBase, HIPPIE, HitPredict, HomoMINT, INstruct, Interactome3D, mentha, MINT, and PINA2 to address format discrepancies and partial dataset overlaps.
Scientific Applications:
- Polyglutamine (polyQ) disease interactome comparison: Comparative analysis of nine polyglutamine disease proteins—androgen receptor (AR), atrophin-1 (ATN1), ataxin 1 (ATXN1), ataxin 2 (ATXN2), ataxin 3 (ATXN3), ataxin 7 (ATXN7), calcium voltage-gated channel subunit alpha1 A (CACNA1A), Huntingtin (HTT), and TATA-binding protein (TBP)—to reveal distinct functional networks and the absence of a single common human interactor across all nine.
- Cross-species PPI comparison: BLAST-enabled comparisons between human and Mus musculus PPIs for AR and TBP to identify overlapping interactors.
Methodology:
Integrates PPI datasets at the species or cross-species level and uses BLAST to identify orthologous proteins for interspecies comparisons.
