ImmuneRegulation

ImmuneRegulation integrates genotypic, gene expression, eQTL, ChIP-seq transcription factor binding, and GWAS catalog data to enable analysis of immune regulatory elements that influence gene expression across human cell types.

Key Features:

  • Extensive data integration: Integrates genotypic information from ~43,000 individuals linked with gene expression data from ~51,000 experiments and hosts >220 million expression quantitative trait loci (eQTL) associations.
  • Transcription factor binding data: Incorporates transcription factor–binding region hits derived from 1,945 ChIP-seq studies totaling ~14 million data points.
  • GWAS catalog integration: Integrates the GWAS catalog containing 67,230 published variant–trait associations.
  • Cross-cohort regulatory association analysis: Enables exploration of associations between queried genes and regulators (cis-eQTLs, trans-eQTLs, and TFs) across multiple cohorts and studies to identify genotype-dependent gene expression variation and inform cohort or cell-type selection.

Scientific Applications:

  • Interpretation of immune regulatory mechanisms: Supports interpretation of how genetic variants and transcription factors regulate gene expression in immune contexts.
  • Predictive modeling of immune responses: Provides comprehensive genetic and transcriptional regulation data for building predictive models of immune responses and disease mechanisms.
  • Cohort and cell-type selection for follow-up studies: Enables selection of cohorts or cell types based on regulatory element associations with genes of interest.

Methodology:

Integration of genotypic data (~43,000 individuals) and gene expression experiments (~51,000), aggregation/hosting of >220 million cis- and trans-eQTL associations, incorporation of TF-binding region hits derived from 1,945 ChIP-seq studies (~14 million data points), and integration of the GWAS catalog with 67,230 variant–trait associations for cross-cohort association analysis of genes with cis-eQTLs, trans-eQTLs, and TFs.
