LINCS Data Portal 2.0

LINCS Data Portal 2.0 provides access to perturbation-response signatures from the Library of Integrated Network-Based Cellular Signatures (LINCS) to support analysis of cellular responses across diverse model systems and assay types.

Key Features:

  • Enhanced Data Architecture and APIs: A revised data architecture and APIs enable programmatic access and integration of LINCS datasets.
  • Curated Metadata Annotations: Enhanced curated metadata annotations support advanced querying and analysis across perturbations, model systems, and signatures.
  • Data Point Level Access: Reprocessing of high-level LINCS datasets enables access and download of signatures at the data-point level, allowing subset retrieval independent of originating source, project, or assay.
  • Signature Search (iLINCS): Integration with the iLINCS platform enables signature-level search to identify conditions that mimic or reverse specific gene set queries.
  • Global Metadata Search: Global metadata search across all annotations supports retrieval of perturbations, model systems, and signatures by specific metadata criteria.

Scientific Applications:

  • Drug discovery: Identification of perturbagens that mimic or reverse disease-relevant gene expression signatures for candidate prioritization.
  • Understanding disease mechanisms: Analysis of perturbation-response signatures to infer pathways and cellular responses underlying disease phenotypes.
  • Exploring gene function: Use of signature comparisons to assess the effects of genetic or chemical perturbations on gene networks and cellular states.
  • Personalized medicine: Granular signature-level queries to support hypothesis generation for context- or patient-specific therapeutic strategies.

Methodology:

Reprocessing of all high-level LINCS datasets to enable data-point-level access; implementation of a revised data architecture and APIs to accommodate diverse perturbations, model systems, and assay types; and incorporation of curated metadata annotations to support advanced querying.
