TCRpMHCmodels
TCRpMHCmodels models T-cell receptor (TCR)–peptide–Major Histocompatibility Complex class I (pMHC class I) complexes to predict three-dimensional structures for analysis of molecular interactions in cellular immune responses.
Key Features:
- Accurate Structural Modeling: Employs comparative modeling to generate three-dimensional models of TCR-pMHC class I complexes.
- Input Specification: Accepts amino acid sequences for all protein chains involved, including TCR alpha chain, TCR beta chain, peptide chain, and MHC chain.
- Automatic Chain Detection: Detects chain types automatically using FASTA headers to assign chain identities.
- Automated Template Selection: Selects optimal structural templates automatically for comparative modeling.
- Reported Accuracy: Achieves a median Cα RMSD (Root Mean Square Deviation) of 2.31 Å and is reported to outperform methods such as TCRFlexDock.
Scientific Applications:
- Insights into Immune Mechanisms: Provides structural models to elucidate molecular determinants of TCR–pMHC recognition and interaction.
- Advancement of Immunotherapies: Supports structural analysis relevant to the design and optimization of T-cell-based immunotherapies.
- Rational Vaccine Design: Informs antigen presentation and immune recognition studies useful for rational vaccine development.
Methodology:
Takes amino acid sequences of TCR alpha, TCR beta, peptide, and MHC chains as input; automatically detects chain types from FASTA headers; selects templates automatically; and applies comparative modeling to predict three-dimensional structures.
