VARIDT

VARIDT catalogs variability of drug transporters, aggregating genetic, epigenetic, tissue- and disease-specific abundance, and exogenous factor data to support analysis of their effects on drug absorption, distribution, and excretion.

Key Features:

  • Comprehensive Data Collection: Consolidates 23,947 DNA methylations, 7,317 noncoding RNA/histone regulations, 1,278 genetic polymorphisms, differential abundance profiles for 257 DTs across 21,781 patients and healthy individuals, expression data for 245 DTs in 67 tissues from human and model organisms, and details on 1,225 exogenous factors affecting activity of 148 DTs.
  • Confirmed Drug Transporters (DTs): Confirms 177 drug transporters based on drugs approved for use and an additional 146 DTs associated with clinical or preclinical drugs.
  • Interconnected Data: Integrates multiple aspects of DT variability to enable mutual connection of genetic, epigenetic, abundance, and exogenous-factor datasets.

Scientific Applications:

  • Clinical Studies: Supports research into how individual differences in DT variability affect drug response and pharmacokinetics.
  • Precision Medicine: Enables consideration of genetic, epigenetic, tissue-specific, and environmental factors for personalized treatment strategies.
  • Disease Etiology and Treatment Optimization: Facilitates identification of variability patterns that may reveal disease mechanisms and inform clinical treatment optimization.

Methodology:

Systematic collection and validation of DT variability data; incorporation of confirmed transporters based on approved and investigational drugs; integrated database structure enabling connections among diverse dataset types.
