ADPriboDB

ADPriboDB curates a comprehensive database of experimentally identified ADP-ribosylated proteins and site-specific ADP-ribosylation information to support research into the roles of ADP-ribosylation in DNA repair, RNA regulation, cell cycle progression, and biomolecular condensate formation.

Key Features:

  • Extensive Repository: Contains 48,346 entries describing 9,097 ADP-ribosylated proteins, including 6,708 proteins newly reported since the original release.
  • Site-Specific Information: Provides site-level ADP-ribosylation annotations for 32,946 entries to enable precise mapping across the proteome.
  • Integration with Interaction Networks: Links ADP-ribosylated substrates to human protein interaction networks and highlights associations relevant to SARS-CoV-2, including the viral macrodomain.
  • Interactive Visualization Tools: Offers visualization of the local context of ADP-ribosylation alongside structural and functional features and other post-translational modifications such as phosphorylation, methylation, and ubiquitination.
  • Curated Data: Entries are curated by at least two independent curators from literature spanning January 1975 to July 2015.

Scientific Applications:

  • Mechanistic Studies: Enables investigation of the specific roles and mechanisms of ADP-ribosylation in cellular processes.
  • Disease Research: Supports analysis of how dysregulation of ADP-ribosylation contributes to diseases such as cancer and neurodegeneration.
  • Viral Infection Studies: Facilitates exploration of the impact of ADP-ribosylation on viral pathogenesis and host responses, including SARS-CoV-2 interactions.

Methodology:

Data derive from proteomics experiments, particularly mass spectrometry identification of ADP-ribosylation sites, and the database integrates those experimental datasets with existing datasets.
