AlzGPS
AlzGPS integrates multi-omics datasets and human protein-protein interactome analyses to identify Alzheimer's disease endophenotype modules, therapeutic targets, and repurposable compounds.
Key Features:
- Multi-Omics Data Integration: Curates over 100 Alzheimer's disease multi-omics datasets encompassing DNA, RNA, protein, and small molecule profiles across early versus late stages and tau versus amyloid endophenotypes.
- Network-Based Analysis: Constructs endophenotype-specific disease modules by integrating multi-omics findings with human protein-protein interactome networks and generates brain-specific neighborhood and mechanism-of-action networks for genes and drugs.
- Drug Repurposing Identification: Employs network proximity analyses to evaluate approximately 3,000 FDA-approved or investigational compounds and curates over 300 literature references supporting repurposable drugs.
- Clinical Trial Integration: Incorporates information from more than 200 ongoing Alzheimer's disease clinical trials, including drug mechanisms and primary targets linked to integrated multi-omics modules.
Scientific Applications:
- Target Identification: Integrates diverse biological data types to prioritize novel therapeutic targets within Alzheimer's disease endophenotypes.
- Therapeutic Development Support: Supports drug repurposing and target prioritization through network-based analyses that link compounds to disease modules.
- Systems Pharmacology: Combines systems pharmacology approaches with multi-omics integration to characterize mechanisms of action for candidate drugs.
Methodology:
Curation of >100 multi-omics datasets (DNA, RNA, protein, small molecules); integration with human protein-protein interactome networks to construct endophenotype-specific disease modules and brain-specific neighborhood and mechanism-of-action networks; application of network proximity analyses to evaluate ~3,000 FDA-approved or investigational compounds and curation of >300 literature references; incorporation of >200 ongoing AD clinical trial records linking drug mechanisms and primary targets to modules.
