ASF-FAST
ASF-FAST performs rapid characterization of African swine fever virus (ASFV) genomes using Oxford Nanopore MinION sequencing and companion analysis software to enable real-time identification and genome-resolved sequencing for outbreak support.
Key Features:
- Rapid Sequencing Capability: Uses the Oxford Nanopore MinION platform to sequence the ~180 kilobase ASFV genome for rapid data generation.
- Real-Time Data Analysis: Processes sequencing output in real time via a companion analysis script to enable immediate identification and characterization of ASFV sequences, with virus-specific reads detectable within approximately 6 minutes of sequencing initiation.
- Host DNA Depletion: Incorporates removal of host-methylated DNA from extracted nucleic acids to enrich virus-specific reads and accelerate detection.
- Comprehensive Genome Resolution: Produces near-complete to complete ASFV genome sequences (up to 100% resolution) in approximately 10 minutes irrespective of starting material.
- Sample Versatility: Generates full ASFV genomes from both cell culture isolates and blood samples from experimentally infected pigs.
- Epidemiological Support: Provides rapid genomic data suitable for timely isolate characterization and outbreak investigation.
Scientific Applications:
- Disease Surveillance and Control: Supplies rapid genomic information for monitoring ASFV spread and informing control measures.
- Diagnostic Laboratories: Enables fast, genome-resolved ASFV identification to support diagnostic workflows and early intervention.
- Research and Development: Facilitates studies of ASFV genetic diversity and evolution relevant to vaccine and countermeasure research.
Methodology:
Processes Oxford Nanopore MinION sequencing output in real time using a companion analysis script to identify ASFV-specific reads and generate genome-resolved sequences within minutes.
