AutoRELACS

AutoRELACS automates generation and analysis of ultra-parallel ChIP-seq data using the Restriction Enzyme-based Labeling of Chromatin in Situ (RELACS) protocol for high-throughput profiling of protein-DNA interactions genome-wide.

Key Features:

  • Automation with Biomek i7: Implements the RELACS protocol on the Biomek i7 liquid handler workstation to automate library preparation.
  • Chromatin barcoding: Applies chromatin barcoding strategies to multiplex samples prior to sequencing.
  • High-throughput ChIP-seq generation: Produces standardized ChIP-seq libraries compatible with high-throughput sequencing.
  • Integration with snakePipes: Is paired with the snakePipes automated computational pipeline for downstream data processing and analysis.
  • Standardization and reduced variability: Automation standardizes sample processing to reduce human error and variability relative to manual RELACS.
  • Compatibility with limited samples: Enables ultra-parallel profiling suited to experiments with limited biological input.
  • Comparable data quality: Generates ChIP-seq data reported to be comparable to manually produced standards.

Scientific Applications:

  • Epigenetic profiling: Enables epigenetic profiling via ChIP-seq measurements of protein-DNA interactions.
  • Genome-wide interaction mapping: Supports genome-wide mapping of protein-DNA interactions using RELACS-derived ChIP-seq.
  • Low-input high-throughput studies: Facilitates high-throughput comparative studies when biological material is limited.

Methodology:

Automated computational analysis of RELACS-derived ChIP-seq data is performed using the snakePipes pipeline to process samples from collection through biological interpretation.
