ComPrAn

ComPrAn analyzes SILAC-labeled complexome profiling data to enable quantitative study of native macromolecular complexes separated by density gradients.

Key Features:

  • Peptide-level analysis: Processes peptide tables in tab-delimited format as input for downstream analysis.
  • Protein-level analysis: Provides normalization and clustering tools for protein-level data.
  • Visualization: Generates dedicated visualization functions and publication-ready figures.
  • 2-state labeled sample compatibility: Supports analysis of 2-state labeled samples separated into fractions for proteomics experiments beyond qDGMS.

Scientific Applications:

  • Macromolecular complex assembly: Inspects assembly and dynamics of native protein and ribonucleoprotein complexes such as the human mitochondrial ribosome.
  • Interacting protein analysis: Identifies proteins interacting with macromolecular assemblies including the small subunit of the cytoplasmic ribosome.
  • Multi-complex profiling: Studies cytoplasmic aminoacyl-tRNA synthetase complex and the mitochondrial PDH complex.
  • Dynamic complex behavior: Accounts for dynamics inherent to biological systems using proteome-wide quantitative and qualitative capability.

Methodology:

Mass spectrometric quantitation via stable isotope labelling with amino acids in cell culture (SILAC), with separation of native complexes by density gradients.
