tadar
tadar standardises the analysis of Differential Allelic Representation (DAR) to identify and mitigate allelic bias artefacts that compromise Differential Expression analysis results in RNA-seq data.
Key Features:
- DAR quantification: Generates an interpretable metric between 0 and 1 for each genetic feature, where 0 indicates identical allelic representation and 1 indicates complete allelic diversity.
- False-positive identification: Flags genetic features prone to false-positive calls in Differential Expression analysis due to DAR artefacts.
- Statistical integration: Enables leveraging of the DAR metric with statistical methods to alleviate the impact of allelic bias artefacts on RNA-seq data.
Scientific Applications:
- RNA-seq quality control: Assesses allelic representation integrity in RNA-seq datasets prior to or alongside Differential Expression analysis.
- Differential Expression validation: Identifies genes or transcripts whose differential expression calls may be confounded by DAR artefacts rather than true biological variation.
Methodology:
The package provides standardised functions for DAR analysis; specific computational algorithms are not detailed in the sources.
