cAb-Rep

cAb-Rep provides a curated database of human B cell immunoglobulin sequence repertoires to support analyses of antibody diversity, somatic hypermutation, isotype distribution, and related repertoire features.

Key Features:

  • Extensive Dataset: The database includes 306 curated B cell repertoires from 121 human donors representing healthy subjects, vaccinated individuals, and those with autoimmune conditions.
  • Heavy and Light Chain Coverage: cAb-Rep comprises 267.9 million V(D)J heavy chain transcripts and 72.9 million VJ light chain transcripts that are full-length or near-full-length.
  • Advanced Annotation: Each transcript is annotated with gene origin, antibody isotype, somatic hypermutations, post-translational modifications, and other relevant characteristics.
  • Error Correction and Deduplication: Redundancy and sequencing errors have been removed from the raw datasets to improve data integrity.
  • FASTA Format: Sequences are stored in FASTA format compatible with repertoire-analysis programs.

Scientific Applications:

  • Antibody Diversity Studies: Explore the breadth and composition of B cell receptor repertoires and their potential to recognize pathogens.
  • Affinity Maturation Analysis: Analyze somatic hypermutation patterns and features relevant to the development of neutralizing antibodies.
  • Algorithm Development: Provide high-quality, annotated datasets for development and benchmarking of immunoglobulin repertoire-analysis algorithms.
  • Prevalence and Reproducibility Studies: Assess prevalence of antibodies with specific genetic signatures and reproducibility of somatic hypermutation patterns.
  • N-glycosylation Frequency Analysis: Support analysis of N-glycosylation frequencies within antibody sequences.

Methodology:

Sequencing was performed using Illumina technology, followed by removal of redundancies and sequencing errors and comprehensive annotation of each transcript for gene origin, isotype, somatic hypermutations, and post-translational modifications.
