CRISP-view

CRISP-view aggregates and curates functional genetic screening datasets from CRISPR/Cas9 and RNA interference (RNAi) experiments to enable analysis of genotype-to-phenotype relationships across phenotypes including in vitro and in vivo cell proliferation and viability, responses to cancer immunotherapy, virus response, and protein expression.

Key Features:

  • Screening technologies: Compiles functional genetic screening datasets generated by CRISPR/Cas9 and RNA interference (RNAi).
  • Phenotype coverage: Includes in vitro and in vivo cell proliferation and viability, responses to cancer immunotherapy, virus response, and protein expression phenotypes.
  • Scope and provenance: Contains 10,321 human samples and 825 mouse samples derived from 167 published studies (as of September 22, 2020).
  • Curation and annotation: Datasets are meticulously curated and annotated.
  • Processing and quality control: Data are processed using the MAGeCK-VISPR analysis pipeline with quality-control metrics maintained across datasets.
  • Data aggregation: Aggregates publicly available genetic screening data to enable cross-study analyses.

Scientific Applications:

  • Functional gene analysis: Enables systematic analysis of gene functions using high-throughput CRISPR and RNAi screens.
  • Genotype-to-phenotype mapping: Supports investigation of genotype-to-phenotype relationships at scale.
  • Identification of phenotype-associated genes: Facilitates discovery of genes associated with proliferation, immunotherapy response, viral response, and protein expression phenotypes.
  • Cross-study comparison: Supports aggregation and comparative analysis across published screening studies.

Methodology:

Datasets are curated and annotated and processed using the MAGeCK-VISPR analysis pipeline with quality-control metrics computed across datasets.
