CRISPRon_off

CRISPRon_off predicts on-target efficacy and potential off-target sites for CRISPR/Cas9 guide RNA (gRNA) design to support selection of gRNAs that maximize editing efficiency while minimizing unintended genomic alterations.

Key Features:

  • Integrated On- and Off-Target Prediction: Combines CRISPRon and CRISPRoff to provide complementary predictions of on-target efficacy and potential off-target sites.
  • Interoperability: Ensures interoperability between CRISPRon and CRISPRoff to streamline the workflow from gRNA selection to validation.
  • Genomic Visualization (IGV): Integrates the Integrative Genomic Viewer (IGV) JavaScript plugin for visualization of gRNA predictions within genomic contexts.

Scientific Applications:

  • Genetic Engineering: Supports design of gRNAs for precise CRISPR/Cas9-mediated genome edits by assessing on-target efficiency and off-target risk.
  • Functional Genomics: Enables selection of gRNAs for loss-of-function or gain-of-function studies with informed off-target considerations.
  • Therapeutic Development: Assists in evaluating gRNA specificity to improve safety and efficacy assessments in CRISPR-based therapeutic research.

Methodology:

Combines CRISPRon and CRISPRoff for on-target and off-target prediction; backend implemented in Python and C; integrates the IGV JavaScript plugin for genomic visualization of gRNA predictions.
