CuticleDB
CuticleDB catalogs and annotates structural proteins of the arthropod cuticle to support studies of cuticle molecular architecture and chitin–protein interactions.
Key Features:
- Extensive Protein Collection: Contains 445 entries, including 370 from insects, 60 from Crustacea, and 15 from Chelicerata.
- Data Sources and Inclusion Criteria: Includes proteins directly sequenced from cuticles, sequences derived from cDNAs that share features with authentic cuticular proteins, and predicted proteins from genomic data.
- Pfam-based Prediction: Predicted cuticular proteins from Drosophila melanogaster and Anopheles gambiae genomes were identified using the Pfam motif PF00379 (chitin-binding domain).
- Motif Analysis: Catalogs motif occurrences and facilitates testing of known and potentially novel motifs specific to arthropod cuticular proteins.
- Genome Annotation Support: Provides curated cuticular protein entries to aid annotation of genomic sequences.
Scientific Applications:
- Genome annotation: Use curated sequences and Pfam PF00379-based predictions to identify and annotate cuticular protein genes in arthropod genomes.
- Molecular structure studies: Investigate relationships between cuticular protein sequences and the helicoidal architecture of the cuticle.
- Chitin–protein interaction research: Analyze chitin-binding motifs and sequence features that mediate protein–chitin interactions.
- Evolutionary and comparative analyses: Compare cuticular protein repertoires across insects, Crustacea, and Chelicerata to study evolution of cuticle composition.
Methodology:
Implemented as a relational database compiling entries from direct protein sequencing, cDNA sequences, and genomic analyses; predicted proteins from Drosophila melanogaster and Anopheles gambiae were identified via Pfam motif PF00379 and motif occurrences are cataloged for analysis.
