CysDB
CysDB consolidates human cysteine chemoproteomics data to provide a curated repository for assessing identification, ligandability (druggability), functional annotation, structural context, and disease-associated variation of cysteine residues across the human proteome.
Key Features:
- Extensive Coverage: Integrates data from nine high-coverage chemoproteomics studies, cataloging identification measures for 62,888 cysteines, representing 24% of human cysteine residues.
- Functional Annotations: Provides detailed annotations on cysteine functionality and ligandability/druggability.
- Disease Relevance and Genetic Variation: Includes information linking cysteine residues to disease relevance and genetic variants.
- Structural Features: Incorporates structural data to provide three-dimensional context for cysteine interactions and function.
- Cross-Study Analysis Capability: Enables integration and comparison of findings across multiple datasets for cross-study analyses.
- Dynamic Growth: Structured to incorporate additional datasets continually to expand characterization of the druggable cysteinome.
Scientific Applications:
- Drug discovery and target prioritization: Supports identification and prioritization of ligandable/druggable cysteine residues for small-molecule and covalent inhibitor development.
- Disease mechanism and variant interpretation: Enables analysis of disease-associated variants and the structural and functional context of cysteines to study disease mechanisms.
- Exploration of untargeted proteome space: Facilitates investigation of therapeutic targets within regions of the human proteome not addressed by existing FDA-approved small molecules.
Methodology:
Aggregates experimental quantitative chemoproteomics data derived from multiple high-coverage studies to capture identification, ligandability, functionality, structural characteristics, and disease-associated annotations of human cysteine residues.
