DDecon

DDecon applies Distributed Deconvolution to periodic line scans from fluorescence images to produce super-resolution measurements of thin filament lengths in striated myofibrils of skeletal and cardiac muscles.

Key Features:

  • Implementation: Provided as an ImageJ plugin for integration with fluorescence-image analysis workflows.
  • Distributed Deconvolution: Employs Distributed Deconvolution (DDecon) analysis to enhance resolution and accuracy of periodic fluorescence signals.
  • Periodic Line Scan Input: Operates on periodic line scans extracted from fluorescence images to analyze regularly spaced structures.
  • Super-Resolution Measurement: Generates high-precision measurements of thin filament lengths within striated myofibrils.
  • Distance Quantification: Quantifies distances of periodically localized fluorescent signals relative to Z- or M-lines and between successive Z- or M-lines.
  • Sample Demonstration: Demonstrated on phalloidin-stained cryosections of mouse skeletal muscle.
  • Applicability Beyond Muscle: Applicable to periodic fluorescence signals in nonmuscle cells as well as skeletal and cardiac muscle.

Scientific Applications:

  • Muscle cytoarchitecture analysis: Measurement of thin filament lengths and spatial relationships within striated myofibrils of skeletal and cardiac muscles.
  • Thin filament regulation studies: Investigation of mechanisms that regulate thin filament lengths across conditions.
  • Developmental and pathophysiological investigations: Assessment of changes in filament length and periodic organization during development or disease states.
  • Genetic perturbation analysis: Quantitative evaluation of structural effects resulting from genetic manipulations.
  • Nonmuscle periodic fluorescence analysis: Analysis of periodically localized fluorescent signals in nonmuscle cells relative to structural landmarks.

Methodology:

Distributed Deconvolution is applied to periodic line scans extracted from fluorescence images; the approach was demonstrated using phalloidin-stained cryosections of mouse skeletal muscle.
