DIANA-miRPath v4.0
DIANA-miRPath v4.0 performs contextual miRNA functional analysis by integrating predicted and experimentally validated miRNA–target interactions to identify enriched pathways and biological terms.
Key Features:
- Contextual Analysis: Customizes target-based miRNA functional analysis according to specific biological or experimental contexts.
- Enrichment Analysis: Performs enrichment on Gene Ontology (GO) terms, KEGG and REACTOME pathways, MSigDB sets, and PFAM domains.
- Integration with Expression Datasets: Integrates bulk and single-cell expression datasets from TCGA, GTEx, and adult/fetal single-cell atlases to assess expression of enriched term components.
- CRISPR Knock-out Screen Analysis: Performs enrichment analyses using CRISPR knock-out screen datasets.
- Custom Data Support: Accepts uploaded custom interaction sets, annotation terms, expression data, and screen datasets.
- Interaction Databases: Uses experimentally supported interactions from DIANA-TarBase v8.0, miRTarBase, and microCLIP cell-type-specific datasets, and in silico predictions from DIANA-microT-CDS and TargetScan.
Scientific Applications:
- Cell-type and tissue-specific miRNA profiling: Enables analysis of miRNA functional roles within specific cell types or tissues.
- Cancer research: Supports discovery of miRNA-associated pathway perturbations using TCGA and related datasets.
- Developmental biology: Facilitates investigation of miRNA function in developmental contexts using adult and fetal single-cell atlases.
- Precision and translational studies: Supports context-specific miRNA analyses relevant to personalized medicine and molecular research.
Methodology:
Uses miRNA interaction sets from DIANA-TarBase v8.0, miRTarBase, and microCLIP cell-type-specific datasets and in silico predictions from DIANA-microT-CDS and TargetScan; performs enrichment analyses on GO, KEGG, REACTOME, MSigDB and PFAM; integrates bulk and single-cell expression datasets from TCGA, GTEx and adult/fetal single-cell atlases; and performs enrichment analysis of CRISPR knock-out screen datasets while supporting custom interaction, term, expression and screen data uploads.
