DIMA 3.0
DIMA 3.0 integrates and predicts protein domain-domain interactions to provide a unified resource for analysis of structural and functional relationships between protein domains.
Key Features:
- Integration of Known Interactions: DIMA incorporates 5,807 structurally validated domain-domain interactions sourced from iPfam and 3did.
- Predictive Computational Methods: Approximately 46,900 predicted domain interactions derived from domain phylogenetic profiling; domain pair exclusion algorithm correlated mutations; and discriminative domain interaction prediction.
- Exclusion of Non-Interacting Pairs: Predicted interactions reported as non-interacting in the Negatome database are filtered out to reduce false positives.
- Domain Interaction Network Calculation: The resource can calculate domain interaction networks tailored to specific domains or entire organisms.
Scientific Applications:
- Protein Structure Prediction: Provides domain-domain interaction evidence to inform modeling of protein tertiary and quaternary structures.
- Functional Genomics: Enables inference of functional relationships between proteins based on domain interaction patterns.
- Drug Discovery and Development: Supplies domain interaction data useful for identifying targets and designing molecules that modulate domain-domain interfaces.
Methodology:
Integration of structurally validated interactions from iPfam and 3did, prediction of approximately 46,900 interactions using domain phylogenetic profiling, domain pair exclusion algorithm correlated mutations, and discriminative domain interaction prediction, filtering with Negatome, and calculation of domain interaction networks.
