DiseaseConnect

DiseaseConnect identifies mechanism-based disease-disease connections by integrating public omics datasets, gene expression data, Genome-Wide Association Studies (GWAS) catalogs, text-mined literature, clinical comorbidity information, and drug-disease interaction compilations.

Key Features:

  • Mechanism-based disease-disease analysis and visualization: Analyzes and visualizes connections between diseases grounded in shared molecular mechanisms and pathological pathways.
  • Omics and literature integration: Integrates omics data and literature-derived evidence, including gene expression datasets and text-mined knowledge, to support mechanistic links.
  • GWAS incorporation: Incorporates Genome-Wide Association Studies (GWAS) catalogs to link genetic associations to disease connectivity.
  • Clinical comorbidity integration: Includes clinical comorbidity information to relate epidemiological co-occurrence with molecular connections.
  • Drug-disease interaction compilation: Compiles known drug-disease interactions to enable mapping between therapeutic agents and mechanism-based disease networks.
  • Mapping of shared mechanisms and pathways: Maps diseases through shared molecular mechanisms and pathological pathways to reveal non-phenotype-based relationships.

Scientific Applications:

  • Discovery of mechanistic disease relationships: Identifies diseases that share molecular mechanisms independent of clinical phenotype grouping.
  • Drug repositioning and therapeutic hypothesis generation: Supports identification of candidate drugs for repositioning by linking drug-disease interactions to shared disease mechanisms.
  • Comorbidity molecular characterization: Enables analysis of comorbidity patterns in the context of underlying molecular pathways.
  • Disease connectivity network construction: Produces disease connectivity networks that integrate genetic, transcriptomic, literature, and clinical data.

Methodology:

Integrates public omics repositories, gene expression datasets, GWAS catalogs, text-mined literature, clinical comorbidity information, and drug-disease interaction compilations to map disease connections via shared molecular mechanisms.
