DynamicProteomics DB

DynamicProteomics DB provides a searchable database of time-lapse yellow fluorescent protein (YFP)-tagged endogenous proteins from the Library of Annotated Reporter Cell-clones (LARC) in living human cells to quantify protein localization and dynamics and their responses to perturbations such as anti-cancer drugs.

Key Features:

  • Protein Tracking: Monitors approximately a thousand proteins tagged with YFP at their endogenous chromosomal locations in living human cells over time.
  • Quantitative Analysis: Produces quantitative fluorescence traces of protein levels in cellular compartments such as the nucleus and cytoplasm derived from image analysis of time-lapse microscopy movies.
  • Sequence Comparison: Provides a sequence analysis tool that compares up to 50 input DNA sequences against all cDNAs in the library.
  • Movie Data and Alignment: Supplies downloadable protein-specific fluorescent movies and aligned dynamics following drug addition, with raw movies available as a benchmark dataset for image-analysis development.
  • Drug-response Dataset: Includes data generated from a human lung carcinoma cell line treated with an anti-cancer drug to enable study of proteome dynamics in response to pharmacological intervention.

Scientific Applications:

  • Cancer Research: Analyze protein behavior and dynamics in response to anti-cancer drugs to investigate mechanisms of drug action and resistance.
  • Proteomics Studies: Track multiple proteins simultaneously within individual cells to study proteomic interactions and pathways.
  • Drug Development: Use detailed temporal protein dynamics to identify potential biomarkers and therapeutic targets.

Methodology:

Image analysis of time-lapse microscopy movies to quantify YFP fluorescence in nucleus and cytoplasm, and sequence-comparison of up to 50 input DNA sequences against the library cDNAs.
