EstimAge

EstimAge computes methylation-based biological age from CpG DNA methylation data using epigenetic clock algorithms to estimate chronological age (years), gestational age (weeks), and cellular age (cell cycles or telomere length in kilobases).

Key Features:

  • Epigenetic clocks: Implements algorithms that use DNA methylation patterns to estimate biological age.
  • CpG-based inputs: Uses methylation states of specific cytosine-guanine dinucleotides (CpGs) as primary predictors.
  • Multiple age metrics: Produces estimates for chronological age in years, gestational age in weeks, and cellular age via cell cycles or telomere length (kilobases).
  • Age acceleration/deceleration detection: Identifies deviations from expected aging patterns indicative of age acceleration or deceleration.
  • Curated clock models: Organizes a curated collection of epigenetic clock models for comparative analysis.
  • Statistical model comparison: Provides statistical tools to compare performance and outputs across different clock models.
  • Model factor annotation: Explicitly addresses CpG site selection, mathematical modeling approaches, tissue specificity, and applicable age ranges across models.
  • Standardization support: Promotes interchangeability and standardization of epigenetic clock algorithms.

Scientific Applications:

  • Aging biology research: Enables comparative evaluation and standardization of epigenetic age assessments in basic and translational studies.
  • Preventive medicine: Detects age acceleration signals that may indicate underlying pathological states prior to clinical manifestation.
  • Diagnostics: Supports use of methylation age as a biomarker for disease-related alterations in biological aging.
  • Forensic science: Provides methylation-based age estimates applicable to forensic age prediction.
  • Cellular aging assessment: Facilitates analysis of cellular age metrics, including relationships with telomere length and cell cycle indicators.

Methodology:

Applies epigenetic clock algorithms to CpG DNA methylation data and performs statistical comparisons across a curated collection of clock models.
