GlycoHunter
GlycoHunter detects and relatively quantifies N-linked glycans labeled by the INLIGHT hydrazide chemistry strategy from MS1 mass spectrometry data.
Key Features:
- Chemical Derivatization Compatibility: Processes N-linked glycans derivatized via hydrazide chemistry using INLIGHT labels NAT (^12C6) and SIL (^13C6).
- Data File Compatibility: Supports MS1 data in imzML and mzXML formats.
- Peak Pair Identification: Identifies MS1 peak pairs corresponding to NAT- and SIL-labeled N-linked glycans.
- Relative Quantification: Computes relative abundances of NAT/SIL-labeled N-linked glycans from identified peak pairs.
- Customizable Search Parameters: Allows modification of search parameters used for MS1 peak-pair detection and quantification.
Scientific Applications:
- Glycomics analysis: Analysis of N-linked glycans in glycomics studies using MS1-based labeling strategies.
- Comparative quantification: Comparative quantification of glycan structures across conditions using NAT/SIL labeling.
- Biomarker discovery: Detection of glycan abundance changes for glycan-based biomarker discovery.
- Disease diagnostics: Characterization of N-linked glycan alterations relevant to disease diagnostics.
- Therapeutic development: Characterization of glycan-mediated biological activities relevant to therapeutic development.
Methodology:
Processes MS1 data from INLIGHT-labeled N-linked glycans derivatized via hydrazide chemistry with NAT (^12C6) and SIL (^13C6), identifies corresponding MS1 peak pairs, and derives relative quantification from those peak pairs.
