gQSPSim
gQSPSim enables development, calibration, and simulation of quantitative systems pharmacology (QSP) models in MATLAB/SimBiology to analyze pharmacokinetic and pharmacodynamic processes such as target-mediated drug disposition and anti-proprotein convertase subtilisin/kexin type 9 (PCSK9) treatment for hypercholesterolemia.
Key Features:
- Model Calibration: Supports calibration using global and local optimization methods for parameter estimation.
- Virtual Subject Development: Enables creation of virtual subjects to characterize variability and uncertainty in biological responses.
- Virtual Population Simulation: Simulates virtual populations under diverse interventions to evaluate population-level outcomes.
- SimBiology Integration: Constructs models using SimBiology components including species, doses, variants, and rules.
- MATLAB Environment: Operates within MATLAB and leverages SimBiology for model formulation and simulation.
- Reproducibility and Portability: Leverages SimBiology components to promote reproducibility, transparency, portability, and reuse in QSP workflows.
Scientific Applications:
- PK/PD modeling: Supports simulation and analysis of pharmacokinetic and pharmacodynamic processes to predict drug behavior and optimize therapeutic strategies.
- Target-mediated drug disposition: Demonstrated with a standard target-mediated drug disposition model.
- PCSK9 inhibition in hypercholesterolemia: Applied to a published anti-proprotein convertase subtilisin/kexin type 9 (PCSK9) model for treatment of hypercholesterolemia.
Methodology:
Operates within the SimBiology environment in MATLAB, using species, doses, variants, and rules for model construction, employing global and local optimization methods for calibration, and providing generation of virtual subjects and simulation of virtual populations under specified interventions.
