GWAS

GWAS prioritizes candidate genes at Parkinson's disease (PD) genome-wide association study loci to aid pinpointing causal genes and variants and support identification of therapeutic targets.

Key Features:

  • Comprehensive data integration: Aggregates and synthesizes data from multiple databases and recent PD GWAS studies to provide contextual evidence for loci.
  • Genome-wide significant signals: Incorporates 92 independent genome-wide significant signals derived from recent PD GWAS, including studies of PD risk, age-at-onset, and progression.
  • Locus gene coverage: Includes information for 2,336 genes located within 1 megabase (Mb) upstream and downstream of each variant to define local candidate sets.
  • Gene prioritization: Enables evaluation and ranking of candidate genes using user-applied self-ranked criteria based on aggregated evidence.

Scientific Applications:

  • Functional follow-up prioritization: Prioritizes genes at PD-associated loci for downstream functional validation experiments.
  • Therapeutic target nomination: Identifies candidate genes that may serve as potential therapeutic targets for PD.
  • Phenotype-specific genetic analysis: Supports investigation of genetic associations related to PD risk, age-at-onset, and progression.
  • Gene-to-variant contextualization: Provides contextual genomic evidence to inform interpretation of GWAS variants within loci.

Methodology:

Aggregates and synthesizes genome-wide significant signals from recent PD GWAS studies, integrates data from multiple databases, and examines genes within a 1 Mb window of each variant while enabling application of self-ranked prioritization criteria.
