HASP
HASP generates comparative structural models of Influenza A hemagglutinin protein sequences to enable three-dimensional interpretation of phylogenetic, biochemical, and immunological data.
Key Features:
- Rosetta-based comparative modeling: Uses the Rosetta modeling suite to produce comparative models of hemagglutinin amino acid sequences.
- Comprehensive sequence set: Provides models for all publicly available Influenza A hemagglutinin amino acid sequences (8,769 as of the publication).
- High structural fidelity: Models exhibit C-alpha root mean square deviations less than 2 Å when compared to crystallographic templates.
- Sequence alignments: Generates sequence alignments corresponding to modeled hemagglutinin sequences for structural mapping.
- Scalable modeling protocols: Modeling protocols are reported to scale with expanding sequencing efforts.
Scientific Applications:
- Structural interpretation of phylogenetic, biochemical, and immunological data: Enables mapping sequence variation onto three-dimensional hemagglutinin structures to interpret phylogenetic, biochemical, and immunological findings.
- Analysis of mutations affecting pathogenicity and species specificity: Supports analysis of hemagglutinin mutations that influence viral pathogenicity and host specificity.
- Comparative analysis with experimental structures: Facilitates comparison between modeled hemagglutinins and related crystallographic templates.
- Large-scale structural analysis: Supports structural analysis across thousands of Influenza A hemagglutinin sequences to track sequence diversity.
Methodology:
HASP uses the Rosetta modeling suite to generate comparative models for publicly available Influenza A hemagglutinin amino acid sequences, produces corresponding sequence alignments, and evaluates model quality by C-alpha root mean square deviation against crystallographic templates.
