HRTBLDb

HRTBLDb catalogs hormone receptor binding loci and associated binding motifs to support analysis of transcriptional regulation by the estrogen receptor (ER), androgen receptor (AR), and glucocorticoid receptor (GR).

Key Features:

  • Receptor coverage: Contains binding loci and motifs for estrogen receptor (ER), androgen receptor (AR), and glucocorticoid receptor (GR).
  • Data integration: Integrates experimental in vivo ChIP-based high-throughput datasets with computationally predicted motifs and cis-regulatory modules.
  • In vitro validation: Includes individual binding sites validated by in vitro experiments, including 114 hormone response elements and 75 co-occurring binding motifs confirmed for co-regulatory action.
  • Dataset composition: Comprises 44,673 binding elements total, including 18,472 binding loci from in vivo ChIP experiments and 26,012 computationally predicted binding motifs.
  • Motif and module annotation: Catalogs computationally predicted binding motifs and cis-regulatory modules associated with transcription factor binding motif co-occurrence within loci.

Scientific Applications:

  • Transcriptional regulation studies: Enables analysis of ER-, AR-, and GR-mediated transcriptional regulation at binding loci and motifs.
  • Regulatory element validation: Supports identification and interpretation of in vitro-validated hormone response elements and co-regulatory motifs.
  • Developmental and tissue biology: Facilitates investigation of hormone receptor roles in cellular differentiation and tissue development in skin, bone, brain, and endocrine tissues.
  • Cis-regulatory analysis: Assists discovery of cis-regulatory modules and motif co-occurrence patterns relevant to hormone receptor binding.

Methodology:

Integration of experimental in vivo ChIP-based high-throughput data, inclusion of in vitro-validated binding sites, and incorporation of computationally predicted binding motifs and cis-regulatory modules.
