ICPLQuant
ICPLQuant processes LC-MALDI mass spectral data from ICPL (Isotope-Coded Protein Label)‑labeled peptides to quantify peptides and integrate quantitative data with MS/MS identifications for detection of regulated proteins.
Key Features:
- ICPL Multiplex Detection and Quantification: Performs MS-level detection and quantification of ICPL-labeled peptides using a dual-unit architecture and proposes specific peptides for subsequent MS/MS analysis.
- Integration with MASCOT for Protein Identification: Integrates quantitative MS data with MASCOT MS/MS identifications to produce protein and peptide lists annotated with quantitative information.
- Efficient Data Processing: Generates lists of differentially regulated peptide precursors to reduce required MS/MS acquisition and manual interpretation.
- Demonstrated Accuracy and Utility: Validated by comparative analysis of mixtures containing Ovalbumin, Horseradish Peroxidase, and Rabbit Albumin spiked into a complex protein background to assess quantification and identification accuracy.
Scientific Applications:
- Quantitative proteomics: Identification and quantification of regulated proteins in complex LC-MALDI datasets labeled with ICPL.
- Biomarker discovery: Detection of differential protein abundance for candidate biomarker selection.
- Disease mechanism and therapeutic target studies: Profiling protein regulation to support elucidation of disease mechanisms and identification of therapeutic targets.
Methodology:
MS-level quantification of ICPL-labeled peptides; selection/proposal of peptides for MS/MS; integration of quantitative data with MASCOT MS/MS for protein identification; generation of lists of differentially regulated peptide precursors; comparative analysis using Ovalbumin, Horseradish Peroxidase, and Rabbit Albumin spiked into a complex protein background.
