Immunolyser

Immunolyser analyzes immunopeptidomic Liquid Chromatography-Mass Spectrometry (LC-MS) datasets to characterize peptides presented by Major Histocompatibility Complex (MHC) molecules and inform studies of antigen processing and presentation.

Key Features:

  • Peptide Length Distribution Analysis: Examines the distribution of peptide lengths within LC-MS immunopeptidomic datasets to assess antigen processing patterns.
  • Peptide Motif Analysis: Identifies recurring amino acid motifs in presented peptides to reveal potential MHC binding preferences.
  • Sequence Clustering: Groups similar peptide sequences to identify related peptide families and repertoire overlap across samples or replicates.
  • Peptide-MHC Binding Affinity Prediction: Predicts binding affinities between peptides and MHC molecules to prioritize candidate antigenic peptides.
  • Source Protein Analysis: Maps identified peptides back to source proteins to link presented peptides with protein origins and degradation pathways.

Scientific Applications:

  • Antigen processing and presentation studies: Characterizes MHC-presented peptides to investigate antigen processing pathways.
  • Candidate antigen identification: Prioritizes peptides with predicted MHC binding for downstream immunogenicity or vaccine research.
  • Comparative immunopeptidomics: Compares peptide repertoires across conditions, samples, or replicates to detect changes in presentation.
  • Peptidome motif discovery: Discovers sequence motifs associated with MHC binding and immune recognition.

Methodology:

Performs peptide length distribution analysis, peptide motif analysis, sequence clustering, peptide-MHC binding affinity prediction, and mapping of peptides to source proteins from immunopeptidomic LC-MS data.
