KairoSight

KairoSight analyzes cardiac optical mapping (optocardiography) data to quantify transmembrane voltage and intracellular calcium dynamics from parameter-sensitive fluorescence dye signals for cardiac electrophysiology studies.

Key Features:

  • Optocardiography signal analysis: Performs quantitative analysis of voltage and intracellular calcium signals derived from parameter-sensitive fluorescence dyes.
  • Comprehensive electrical and calcium dynamics quantification: Extracts and measures electrical activity and calcium transients to characterize cardiac electrophysiological properties.
  • Large dataset handling: Processes large optical mapping datasets to enable detailed spatial and temporal analyses.
  • Multispecies experimental support: Demonstrated with datasets from Langendorff-perfused rat, guinea pig, and swine hearts and human induced pluripotent stem cell-derived cardiomyocytes (hiPSC-CM).
  • Fluorescence dye compatibility: Supports common voltage and calcium indicators including RH237, Rhod-2 AM, FluoVolt, and Fluo-4 AM.

Scientific Applications:

  • Pharmacological response analysis: Enables assessment of cardiac responses to agents such as ryanodine and isoproterenol by analyzing changes in voltage and calcium signals.
  • Regional electrophysiology and injury studies: Facilitates investigation of spatial heterogeneity and regional differences in tissue properties following ablation injuries.
  • Comparative and translational studies: Supports cross-species and model comparisons, including Langendorff-perfused hearts and hiPSC-CM, for translational cardiac research.

Methodology:

Implemented in Python and processes optical mapping fluorescence datasets to extract and quantify transmembrane voltage and intracellular calcium signals.
