Leonardo
Leonardo corrects sample-induced artifacts in selective plane illumination microscopy (SPIM) images through de-striping and image reconstruction modules.
Key Features:
- DeStripe: Removes stripe artifacts in SPIM caused by light absorption while preserving fine sample structures.
- Fuse: Reconstructs a single high-quality image from dual-sided illumination and/or dual-sided detection while eliminating blur and optical distortions caused by light scattering and refraction.
Scientific Applications:
- Live imaging of sensitive specimens: Validates performance on translucent embryonic and optically opaque larval zebrafish in minimally invasive experiments.
- Large cleared tissue imaging: Validates performance on cleared mouse samples up to two centimeters in size.
- Morphogenesis and function studies: Enables extended-period volumetric imaging of biological specimens by addressing light absorption, scattering, and refraction artifacts.
Methodology:
The toolbox uses two modules: DeStripe for removing absorption-induced stripe artifacts, and Fuse for reconstructing images from dual-sided illumination and/or detection to eliminate scattering and refraction-induced blur and distortions.
