MAGI-MS
MAGI-MS identifies genetic modules by integrating multiple seed genes with gene co-expression and protein-protein interaction networks to detect clusters of highly co-expressed genes associated with specific biological pathways.
Key Features:
- Multiple Seed Gene Integration: Allows specification of a disease pathway by using multiple seed genes that function within the same molecular mechanism to increase module specificity.
- Co-expression and Protein-Interaction Integration: Leverages gene co-expression data and protein-protein interaction networks to identify clusters of highly co-expressed genes.
- Pathway-Specific Module Discovery: Focuses on pairs or sets of seed genes to uncover modules enriched for functions such as chemical synaptic transmission and glutamatergic synapse activity.
- Phenotype-Relevant Module Identification: Detects gene clusters that participate in common biological pathways relevant to specific phenotypes.
- Analysis of Complex Genetic Architecture: Supports investigation of disorders arising from interactions among multiple genetic components.
Scientific Applications:
- Complex disorder module discovery: Identification of gene modules underlying complex, polygenic phenotypes.
- Neurodevelopmental disorder pathway analysis: Dissection of molecular mechanisms in disorders such as epilepsy, including synaptic transmission and glutamatergic synapse pathways.
- Pathway-level interpretation: Identification of modules and enriched pathway components to inform studies of molecular mechanisms and potential therapeutic targets.
Methodology:
Constructs genetic modules from co-expressed genes by leveraging co-expression data and protein-interaction networks, using multiple seed genes to refine identification of relevant biological pathways.
