MagMet automates targeted 1D ^1H NMR metabolomics processing and quantification of human serum and plasma to detect and measure metabolites, including markers of inborn errors of metabolism (IEM).

  • Spectral parameter utilization: Identifies and quantifies metabolites based on peak positions, linewidth information, and J-couplings from experimentally acquired spectra.
  • Linewidth adjustment across field strengths: Adjusts linewidth to ensure consistent quantification across different NMR instrument field strengths.
  • High-throughput performance: Detects and quantifies 58 serum/plasma metabolites in approximately 2.6 minutes per spectrum when processing datasets of 50-100 spectra.

Methodology:

An optimized algorithm integrates automatic Fourier transformation, phase correction, baseline optimization, chemical shift referencing, water signal removal, peak picking and alignment, and linewidth adjustment, using a reference spectral NMR library of 85 serum/plasma compounds to identify and quantify metabolites based on peak positions, linewidths, and J‑couplings.
