MTSviewer

MTSviewer enables visualization and analysis of mitochondrial targeting sequences (MTS), cleavage sites, and mutations mapped onto protein structures to study mitochondrial protein import and processing.

Key Features:

  • Integration of predictions and experimental data: Combines predictions of MTSs and cleavage sites with N-terminomics data, genetic variants, and pathogenicity assessments.
  • Structural mapping: Maps sequence features and variants onto AlphaFold models for human and yeast mitochondrial proteomes.
  • Proteome-wide profiling: Profiles MTS-containing proteins across human and yeast mitochondrial proteomes to survey the mutational and structural landscape.
  • Case studies: Provides example analyses that investigate roles of MTSs and their implications in mitochondrial dysfunction, including links to neurodegenerative disorders and cardiovascular defects.

Scientific Applications:

  • Study of protein import and homeostasis: Analyze how MTSs and cleavage events contribute to mitochondrial protein import and mitochondrial homeostasis.
  • Characterization of MTS types: Investigate conventional N-terminal MTSs and internal MTSs that act as import regulators or secondary cleavage sites within proteins.
  • Variant pathogenicity assessment: Integrate genetic variants with structural context to evaluate the potential pathogenicity of mutations within MTSs and adjacent regions.
  • Disease mechanism investigation: Explore molecular links between MTS alterations and mitochondrial dysfunction in conditions such as neurodegenerative disorders and cardiovascular defects.

Methodology:

Integrates MTS and cleavage-site predictions with N-terminomics data, maps genetic variants and pathogenicity assessments onto AlphaFold structural models, and profiles MTS-containing proteins across human and yeast mitochondrial proteomes.
