multiSero

multiSero measures antibody responses to multiple antigens using a multiplex enzyme-linked immunosorbent assay (ELISA) platform for serosurveillance.

Key Features:

  • Multiplexing Capability: Concurrent assessment of antibodies to multiple proteins within a 96-well ELISA array.
  • Automated Imaging and Analysis: Automated imaging using an open-source plate reader coupled to an analysis pipeline that quantifies optical densities for each protein in the array.

Scientific Applications:

  • SARS-CoV-2 Seropositivity Validation: Validated on human sera against SARS-CoV-2 antigens with sensitivity 0.978, specificity 0.977, positive predictive value 0.978, and negative predictive value 0.977.
  • Correlation with Commercial Tests: Demonstrates a strong correlation with commercially available SARS-CoV-2 antibody tests.
  • Vaccine Response Monitoring: Tracks antigen-specific changes in antibody titer dynamics post-vaccination.
  • Multiplex Serosurveillance: Profiles immune responses to diverse antigens to support serosurveillance across multiple pathogens.

Methodology:

Automated imaging with an open-source plate reader and an automated analysis pipeline that measures optical densities per protein in the 96-well multiplex ELISA array.
