NbThermo

NbThermo compiles curated thermodynamic and sequence data for nanobodies (Nbs), single-domain camelid heavy-chain antibody fragments, to support analysis and prediction of nanobody thermostability.

Key Features:

  • Extensive Data Collection: Contains curated data on 564 nanobodies, including melting temperatures (Tm) and amino acid sequences derived from the literature.
  • Focus on Thermostability: Emphasizes melting temperature (Tm) as a primary metric for assessing nanobody stability alongside sequence information.
  • Source Organism Insights: Includes nanobodies sourced from llama and camel and reports similar Tm distributions between these species.
  • Exploratory Research Support: Enables investigation of structural determinants of thermostability, with initial analyses indicating framework sequences do not significantly differ between low- and high-Tm groups, implicating variable loops.
  • Algorithm Development: Provides a dataset suitable for developing and validating computational models to predict nanobody melting temperatures.

Scientific Applications:

  • Predictive Modeling: Development of computational models to predict nanobody melting temperatures from sequence data.
  • Protein Engineering: Design of nanobodies with enhanced stability for therapeutic, diagnostic, and industrial applications.
  • Structural Analysis: Investigation of structural features, particularly variable loops and framework regions, that contribute to thermostability.

Methodology:

Data were gathered through an extensive literature search and manually curated to compile nanobody melting temperatures and amino acid sequences.
