PASA
PASA maps antigen-specific peptides identified in high-resolution proteomic mass spectra to B-cell receptor sequences from BCR-Seq to characterize the serum antibody repertoire.
Key Features:
- Quantitative and Qualitative Analysis: Enables both quantitative and qualitative measurements of the serum antibody repertoire using high-resolution proteomic data.
- Data Integration: Integrates proteomics data with next-generation sequencing (BCR-Seq) data to map antigen-specific peptides to individual B-cell clones.
- Input Requirements: Accepts proteomics spectral files in '.raw' format and requires at least two technical replicates of each fraction (elution and flow-through).
Scientific Applications:
- B Cell Development and Differentiation: Supports analysis of B cell development and differentiation by profiling antibody repertoires.
- Vaccine Development: Facilitates investigation of vaccine-induced humoral responses through serum antibody characterization.
- Immunodiagnostic Discovery: Aids discovery of immunodiagnostic markers by identifying antigen-associated antibody peptides and clones.
- Monoclonal Antibody Engineering: Enables identification of antigen-binding B-cell clones for monoclonal antibody engineering.
Methodology:
Maps peptides derived from antigen-specific antibodies detected in high-resolution proteomic data to corresponding antibody sequences from BCR-Seq data.
