PepDistiller refines peptide-spectrum match validation to improve peptide identification accuracy and false discovery rate (FDR) estimation in mass spectrometry-based proteomics.

  • Incorporation of Tryptic Termini: Considers the number of tryptic termini in peptides to support validation of semitryptic search results.
  • Refined False Discovery Rate (FDR) Calculation: Implements an enhanced FDR calculation that yields more accurate estimations compared with traditional methods such as the percentage of incorrect target (PIT) fixed method used in MASCOT Percolator.
  • Improved Sensitivity and Accuracy: Identifies approximately 7% more peptides than MASCOT Percolator and reports lower FDR estimations.
