PhenoM

PhenoM profiles phenotypic consequences of temperature-sensitive (ts) alleles of essential Saccharomyces cerevisiae genes by quantifying single-cell morphology from high-throughput microscopy to enable analysis of essential gene functions.

Key Features:

  • Dataset size: Quantitative measurements from 1,909,914 cells and 78,194 morphological images derived from high-throughput microscopy.
  • Mutant collection: Data cover 775 temperature-sensitive (ts) mutants representing 491 essential Saccharomyces cerevisiae genes.
  • Temperature conditions: Measurements obtained at permissive (26°C) and restrictive (32°C) temperatures.
  • Single-cell morphology: Quantified single-cell morphological measurements across six subcellular compartments or structures.
  • PhenoBlast module: Enables phenotypic comparisons between different mutant strains.
  • Gene Ontology module: Performs functional enrichment analysis on gene sets with similar morphological changes.
  • Data mining and visualization: Supports storage, retrieval, visualization, and data-mining of single-cell morphological measurements and raw images.

Scientific Applications:

  • Essential gene phenotyping: Study phenotypic consequences of essential gene mutations using ts alleles under permissive and restrictive temperatures.
  • Phenotype comparison: Identify phenotypic similarity and differences among mutant strains using PhenoBlast.
  • Functional enrichment: Link morphological phenotypes to biological processes and components via Gene Ontology enrichment.
  • Compartment-specific analysis: Examine how essential gene perturbations impact six distinct subcellular compartments.
  • Quantitative single-cell analysis: Perform large-scale quantitative analysis of cellular morphology across genotypes and temperatures.

Methodology:

High-throughput microscopy produced images and quantitative single-cell morphological measurements at 26°C and 32°C; computational tools include the PhenoBlast module for phenotypic comparisons and a Gene Ontology module for functional enrichment analysis.
