PhysiPKPD

PhysiPKPD integrates pharmacokinetics (PK) and pharmacodynamics (PD) into the PhysiCell physics-based cell simulator to model the distribution, action, and tissue-level effects of therapeutic agents.

Key Features:

  • Standardized framework: Provides a consistent representation of PK and PD within PhysiCell models to improve reproducibility.
  • PK model integration: Implements models describing absorption, distribution, metabolism, and excretion of therapeutic agents.
  • PD model integration: Implements models characterizing biochemical effects of agents on cellular processes.
  • Physics-based tissue simulation: Leverages PhysiCell's physics-based cell simulation to represent tissue-scale transport and cellular interactions with drugs.
  • Parameter customization: Supports specification of PKPD parameters to match experimental conditions or therapeutic scenarios.

Scientific Applications:

  • In silico modeling: Simulates drug behavior and cellular responses within virtual tissue models.
  • Therapeutic optimization: Enables exploration and optimization of dosing regimens and therapeutic strategies using PKPD dynamics.
  • Interdisciplinary research: Provides a computational platform for computational biologists, pharmacologists, and clinicians to study drug–tissue interactions.

Methodology:

Uses PhysiCell's physics-based cell simulation and integrates PK models (absorption, distribution, metabolism, excretion) with PD models describing biochemical effects on cellular processes to simulate drug distribution and action in tissues.
