PolyAtailor
PolyAtailor detects and characterizes poly(A) tails from short-read and long-read sequencing data to enable analysis of polyadenylation and mRNA post-transcriptional regulation.
Key Features:
- Unified framework: Identifies and analyzes poly(A) tails across short-read and long-read sequencing technologies.
- Tail_map: Maps poly(A) tails within sequencing data and supports mapping with or without a reference genome.
- Tail_scan: Scans reads to identify all potential poly(A) tails and reports tail position, length, sequence, and type.
- Differential poly(A) length analysis: Compares poly(A) tail length distributions across conditions or developmental stages.
- Poly(A) site identification and annotation: Detects and annotates polyadenylation sites in sequencing data.
- Base-composition statistics and visualization: Provides statistical and visual analysis of nucleotide composition within poly(A) tails.
- High sensitivity and accuracy: Demonstrated higher sensitivity and accuracy measuring poly(A) tail length on HeLa and Arabidopsis datasets compared with FLAMAnalysis, FLEPSeq, and PAIsoSeqAnalysis.
Scientific Applications:
- Post-transcriptional regulation: Enables study of gene expression regulation via mRNA polyadenylation and tail length dynamics.
- Developmental and comparative transcriptomics: Profiles poly(A) tails across developmental stages and different organisms such as HeLa cells and Arabidopsis.
- Functional polyadenylation studies: Provides data to investigate functional implications of polyadenylation dynamics in molecular biology, genetics, and transcriptomics.
Methodology:
Processes short-read and long-read sequencing data using advanced algorithms; Tail_map maps poly(A) tails with or without a reference genome; Tail_scan scans reads to identify all potential poly(A) tails and outputs tail position, length, sequence, and type.
