ProSight PTM
ProSight PTM identifies proteoforms by probability-based interpretation of absolute fragment ion masses and sequence tags to detect and localize post-translational modifications.
Key Features:
- Retriever Algorithm: Performs probability-based identifications using absolute fragment ion masses with optional sequence-tag compilation for enhanced accuracy.
- MySQL Protein Databases: Stores and queries protein sequence and proteoform information using MySQL database integration.
- File/Data Manager: Manages and organizes experimental mass spectrometry data files for analysis.
- Databases for Known and Putative Proteins: Supports searches against databases of known protein forms and putative variants including prior or predicted PTM knowledge.
Scientific Applications:
- Large-Scale Proteome Projects: Identification of a 36-kDa yeast protein from processed cell extract using a quadrupole-FT hybrid mass spectrometer, with localization of an acrylamide-induced alkylation modification on glyceraldehyde-3-phosphate dehydrogenase.
Methodology:
ProSight PTM employs a probability-based identification approach using absolute fragment ion masses derived from mass spectrometry data and can automatically compile sequence tags for improved accuracy.
