PROSITE
PROSITE provides a database of conserved protein patterns, profiles, and ProRules for identifying and annotating protein domains, families, and functional sites in protein sequences.
Key Features:
- Patterns: A collection of 1,308 consensus sequence patterns for recognition of conserved motifs and functional sites.
- Profiles: A set of 1,039 profiles for detecting protein families and domains across sequence space.
- ProRules integration: 1,041 ProRules that add rule-based context to signatures to increase specificity and highlight functionally or structurally critical amino acids.
- PROSITE signatures: Use of signature-based detection (patterns and profiles) for annotation of protein features.
- UniProtKB/Swiss-Prot annotation: Application of PROSITE signatures and ProRules to annotate domains and features within UniProtKB/Swiss-Prot entries.
- Whole-proteome annotation: Support for comprehensive whole-proteome annotation enabled by developments in the resource.
- Filtering options: A suite of filtering options to tailor searches and refine annotations.
- Release statistics: Release 20.85 (30 August 2012) reporting the above counts of patterns, profiles, and ProRules.
Scientific Applications:
- Domain and family annotation: Identification and classification of protein domains and families in sequence datasets.
- Functional site identification: Detection of conserved functional residues and motifs within proteins.
- Whole-proteome analysis: Genome-scale annotation of protein features across complete proteomes.
- Protein analysis and functional genomics: Support for studies linking sequence features to protein function and genomics analyses.
- Targeted searches: Focused identification of proteins or sites using signature matches combined with filtering rules.
Methodology:
Annotation is performed by applying PROSITE signatures (patterns and profiles) in conjunction with ProRules to UniProtKB/Swiss-Prot entries, with support for whole-proteome annotation and configurable filtering options.
