Reactome-IDG

Reactome-IDG integrates understudied "dark" proteins into Reactome's manually curated biological pathways to elucidate their molecular functions and therapeutic potential.

Key Features:

  • Pathway integration: Incorporates dark proteins into Reactome's manually curated pathways to place them in established biological contexts.
  • Visualization: Represents pathway information using Systems Biology Graph Notation (SBGN) diagrams and a simplified Functional Interaction (FI) network view.
  • Tissue-specific expression overlay: Maps tissue-specific gene or protein expression data onto pathway and network representations.
  • Drug-target interaction mapping: Links drug-target interaction data to proteins within pathways and networks.
  • Pairwise relationship integration: Incorporates protein or gene pairwise relationships to support pathway-level functional inference.

Scientific Applications:

  • Function prediction: Supports prediction of molecular functions for understudied "dark" proteins by contextualizing them in curated pathways and interaction networks.
  • Target identification: Aids identification of candidate therapeutic targets by linking dark proteins to drug-target interactions and pathway roles.
  • Mechanistic insight: Facilitates elucidation of molecular mechanisms underlying phenotypes and diseases through integrated pathway, expression, and interaction data.

Methodology:

Integration of dark proteins into Reactome's manually curated pathways, representation with Systems Biology Graph Notation (SBGN) diagrams and a Functional Interaction (FI) network view, overlay of tissue-specific gene or protein expression data, incorporation of protein/gene pairwise relationships, and mapping of drug–target interactions.
