ReFRAME: Drug Repurposing Compound Library for Neglected Diseases

ReFRAME aggregates 12,000 compounds with prior human use to enable systematic drug repurposing and accelerate identification of therapeutic candidates for neglected diseases.

  • Curated Compound Library: Integrates 12,000 compounds from Clarivate Integrity, GVK Excelra GoStar, and Citeline Pharmaprojects, supplemented by patent mining of small molecules previously administered to humans.
  • Human Safety Profiling: Prioritizes compounds with established clinical testing histories to facilitate rapid translational advancement.
  • Chemical Diversity Coverage: Provides structurally diverse small molecules suitable for high-throughput phenotypic and target-based screening.

Scientific Applications:

  • Antiparasitic Drug Discovery: Screening against Cryptosporidium spp. identified VB-201 and an analog of ASP-7962, both previously tested in humans and validated in animal models at clinically relevant doses.
  • Neglected Tropical Disease Research: Enables rapid evaluation of approved or clinically tested compounds for alternative therapeutic indications.

Methodology:

Compounds with documented human administration were aggregated from commercial pharmacological databases and patent-mined sources to create a chemically diverse, clinically informed library. Systematic screening against disease-relevant biological targets or organisms supports identification of repurposable candidates with established safety data.
