scDrugPrio

scDrugPrio prioritizes and repurposes drugs for immune-mediated inflammatory diseases (IMIDs) by constructing and interrogating single-cell RNA sequencing (scRNA-seq)-derived network models to predict drug responses.

Key Features:

  • Network Model Construction: Builds network models by integrating cell type-specific expression changes, altered cellular interactions, and pharmacological properties from scRNA-seq data.
  • Drug Prioritization and Repurposing: Systematically ranks thousands of drugs and predicts drug responses using the constructed network models to identify approved and novel therapeutic candidates.
  • Validation Across Models: Initially developed in a mouse model of antigen-induced arthritis with enhanced precision and recall for known drugs and further validated with in vitro, in vivo, and in silico studies.
  • Application to Multiple Diseases: Applied to multiple sclerosis, Crohn's disease, and psoriatic arthritis to prioritize relevant drugs across these IMIDs.
  • Personalized Medicine Potential: Analyzes scRNA-seq from individual Crohn's disease patients to reveal interindividual variability in drug predictions that correlate with patient-specific responses to anti-TNF therapy.

Scientific Applications:

  • Precision Medicine: Enables personalized network-based drug screening from scRNA-seq profiles to support tailored therapeutic strategies.
  • Research in IMIDs: Facilitates dissection of cellular and molecular heterogeneity and identification of therapeutic targets in immune-mediated inflammatory diseases.
  • Drug Discovery and Repurposing: Accelerates identification and prioritization of candidate drugs for repurposing and development based on network-model predictions.

Methodology:

Analyzes scRNA-seq data from disease models or patient samples and constructs network models by integrating cell type-specific expression changes, altered cellular interactions, and known pharmacological properties to systematically evaluate drug efficacy across cellular contexts; predictions were validated with in vitro, in vivo, and in silico studies.
