sciMET-cap
sciMET-cap leverages target enrichment with sciMET to enable high-throughput single-cell DNA methylation analysis with substantially fewer sequence reads per cell while retaining sufficient information for cell type assignment and genome-wide differentially methylated region calling.
Key Features:
- Target enrichment: Reduces sequencing burden per cell by capturing targeted regions rather than performing whole-genome single-cell DNA methylation sequencing.
- Cell type assignment: Enables accurate cell type identification using substantially fewer sequence reads compared to genome-wide sciMET.
- Genome-wide DMR calling: Accumulated off-target coverage supports genome-wide differentially methylated region calling for clusters with as few as 115 cells.
Scientific Applications:
- Human PBMC characterization: sciMET-cap has been applied to characterize DNA methylation patterns in human peripheral blood mononuclear cells.
- Human brain tissue analysis: sciMET-cap has been applied to characterize DNA methylation in human middle frontal gyrus brain tissue.
- Epigenomic studies: Supports investigation of DNA methylation regulatory roles in development, disease, and other biological processes at single-cell resolution with reduced sequencing requirements.
Methodology:
sciMET-cap builds upon the sciMET assay by incorporating target enrichment to reduce sequencing burden while maintaining analytical capabilities for cell type assignment and differentially methylated region detection through accumulated off-target coverage.
