SIGURD
SIGURD enables clonal analysis of single-cell RNA-sequencing data through quantification of clones, functional analysis, and association of somatic and mitochondrial variants.
Key Features:
- Clonal Quantification: Quantifies clones by leveraging both somatic and mitochondrial variants from scRNA-seq data.
- Functional Analysis: Associates clones with cell populations and detects differentially expressed genes across clones after clonal detection.
- Variant Association: Associates somatic and mitochondrial variants to characterize clonality.
Scientific Applications:
- Myeloproliferative Neoplasm Research: Analyzes single-cell data of colony-forming cells derived from patients with myeloproliferative neoplasms to demonstrate clonal characterization.
- Clone Identification: Uses mitochondrial variants as natural barcodes for clone identification when somatic variant capture is limited by transcript sparsity or protocol bias.
Methodology:
SIGURD is implemented as an R-based pipeline.
